Descripción
BLNK (B-cell linker) is an adaptor protein primarily expressed in B cells, where it plays a crucial role in regulating B cell receptor (BCR) signaling. It functions by linking various signaling molecules, which is critical for the proper activation, differentiation, and survival of B cells. Upon BCR engagement, BLNK is phosphorylated, leading to the recruitment of other signaling proteins, including those involved in the activation of key pathways such as the phosphoinositide 3-kinase (PI3K) and mitogen-activated protein kinase (MAPK) pathways. This allows BLNK to help orchestrate B cell activation and the immune response. It is also involved in regulating B cell proliferation and the transition from the naive to the activated state, essential for effective immune responses. Mutations in BLNK can result in immune dysfunction and are associated with immune-related diseases, including lymphoid malignancies. Understanding its role in B cell signaling can help inform therapies for diseases involving B cells, such as autoimmune diseases and B-cell malignancies.
