Description
The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. The low intrinsic GTPase activity of RAB43 is activated by USP6NL. Involved in retrograde transport from the endocytic pathway to the Golgi apparatus. Involved in the transport of Shiga toxin from early and recycling endosomes to the trans-Golgi network. Required for the structural integrity of the Golgi complex. Plays a role in the maturation of phagosomes that engulf pathogens, such as S.aureus and M.tuberculosis.

Descripción
RAB43, member RAS oncogene family (RAB43) is a member of the RAB family of small GTPases that plays a critical role in regulating vesicle trafficking within the cell. RAB43 is involved in the transport of vesicles between the Golgi apparatus and the endosomal system, helping to mediate protein sorting and trafficking processes essential for cellular homeostasis. This protein is particularly important for regulating the recycling and degradation of membrane proteins, which affects cellular communication and signaling. RAB43 is also implicated in the regulation of receptor endocytosis, a process that is vital for maintaining the responsiveness of cells to external stimuli. In neurons, RAB43 contributes to the movement of synaptic vesicles, ensuring the proper release and recycling of neurotransmitters. Given its role in vesicular transport, RAB43 has been linked to a variety of diseases, including cancer, where disruptions in vesicle trafficking can lead to alterations in receptor signaling pathways and contribute to tumor progression.
