Description
ALDH1L1 catalyzes the conversion of 10-formyltetrahydrofolate, NADP, and water to tetrahydrofolate, NADPH, and carbon dioxide. The encoded protein belongs to the aldehyde dehydrogenase family and is responsible for formate oxidation in vivo. Deficiencies in this gene can result in an accumulation of formate and subsequent methanol poisoning.

Descripción
ALDH1L1, also known as 10-formyltetrahydrofolate dehydrogenase, is a cytosolic enzyme involved in folate metabolism, where it catalyzes the conversion of 10-formyltetrahydrofolate to tetrahydrofolate, a key process in one-carbon metabolism. This enzyme plays a significant role in nucleotide biosynthesis, DNA methylation, and cellular proliferation by regulating the folate cycle and one-carbon pools. ALDH1L1 is highly expressed in the liver and brain, where it supports metabolic processes and detoxification. It functions as a tumor suppressor, as its loss or downregulation is associated with various cancers, including liver, breast, and lung cancers, due to impaired folate metabolism and increased cellular proliferation. ALDH1L1 also contributes to cellular detoxification by reducing reactive aldehydes and maintaining redox balance. Emerging studies highlight its potential as a biomarker for cancer prognosis and as a therapeutic target for restoring normal folate metabolism in tumor cells.
