Descripción
AKAP11, also referred to as AKAP220, is a large cytosolic scaffold protein that binds PKA, phosphodiesterase-4D (PDE4D), and other signaling enzymes, coordinating cAMP and PKA signaling pathways in diverse cellular contexts. AKAP11 is highly expressed in neurons and spermatozoa, where it regulates PKA-dependent signaling for processes like synaptic plasticity, sperm motility, and energy metabolism. AKAP11 forms complexes with PDE4D, ensuring spatial and temporal regulation of cAMP levels, which is critical for cell signaling specificity. Dysregulation of AKAP11 has been linked to infertility, as its loss impairs sperm flagellar movement and mitochondrial ATP production. In neurons, AKAP11 modulates cAMP signaling involved in synaptic transmission, learning, and memory. Studies have also identified AKAP11 as a target in cancer biology, where its misregulation influences cell proliferation, migration, and anchoring of signaling components that regulate growth pathways like MAPK and AMPK.
