Background

HTR3C is a subunit that combines with HTR3A to form functional heteromeric 5-HT3 ion channels, which mediate serotonin-induced depolarization in neurons. Unlike HTR3A, HTR3C alone cannot form functional receptors but instead modifies the biophysical properties of the channel when co-assembled with HTR3A. HTR3C is expressed in the gastrointestinal tract and regions of the central nervous system, where it contributes to the regulation of nausea, pain signaling, and gastrointestinal motility. Studies suggest that genetic variations in the HTR3C gene influence susceptibility to functional gastrointestinal disorders, including IBS and visceral hypersensitivity. Although HTR3C's expression is lower than other 5-HT3 receptor subunits, its modulatory role remains significant for understanding serotonin's effects on neurotransmission and peripheral tissues. The HTR3C subunit broadens the diversity of receptor functions, influencing pharmacological responses to 5-HT3 antagonists like ondansetron and granisetron, which are widely used to manage nausea and vomiting.
