Background:

Aminoterminal propeptide of type I procollagen (PINP) is a metabolite of type I collagen that is released into the bloodstream during the process of new bone formation. Its levels directly correlate with the rate at which bone turnover occurs. In conditions affecting bone metabolism such as osteoporosis, PINP serves as a sensitive indicator to identify individuals with heightened bone turnover and consequently, a greater risk of fractures. PINP proves invaluable in monitoring the efficacy of both anti-resorptive and anabolic treatments. Type I collagen, comprising 90% of bone proteins, is initially synthesized as type I procollagen. During extracellular processing, there's a cleavage leading to the formation of amino terminal [N-terminal propeptide of type I collagen (P1NP)] and carboxy terminal propeptide (P1CP). These propeptides circulate in the bloodstream, serving as markers of bone formation. Unlike serum P1CP, which exists as a single protein, P1NP circulates in various forms, including intact trimeric P1NP, monomers, and several fragments.

  • Domains: It is the N-terminal propeptide of procollagen type I, which is cleaved off during the maturation of procollagen into collagen.
