APOH - Apolipoprotein H | Elisa - Clia - Antibody - Protein

Family main features

Background:

Apolipoprotein H (APOH), also known as beta-2-glycoprotein I (β2GPI), is a multifunctional protein While primarily known for its function in lipid metabolism and regulation of lipid transport in the blood, it is also involved in other physiological processes. APOH is one of the proteins associated with lipoproteins, which are particles responsible for transporting lipids in the blood. It helps stabilize and regulate the structure of these particles, facilitating the transport of lipids such as cholesterol and triglycerides through the bloodstream. APOH has anticoagulant and antithrombotic properties. It has been shown to interfere with blood clotting by inhibiting the activity of certain coagulation factors, such as thrombin, thereby helping prevent the formation of unwanted blood clots. APOH may also play a role in modulating the immune system. It has been shown to interact with antibodies and other immune system proteins, although its exact function in this context is not fully understood.APOH has also been studied in relation to various diseases, including cardiovascular diseases, thrombosis, autoimmune diseases, and pregnancy complications. Anomalies in APOH levels or function are believed to contribute to the development or progression of these conditions.

Protein Structure:

  • Primary Structure: APOH consists of approximately 345 amino acids.
  • Secondary Structure: It features several domains that are rich in beta sheets.
  • Tertiary Structure: The protein has a multi-domain structure, including five distinct short consensus repeat (SCR) domains, which contribute to its ability to interact with various molecules.
  • Quaternary Structure: APOH can form homodimers, particularly when binding to anionic phospholipids.

Classification and Subtypes:

APOH belongs to the apolipoprotein family, which includes proteins essential for lipid transport and metabolism. It is a distinct member without known subtypes or significant isoforms resulting from alternative splicing.

Function and Biological Significance:

  • Lipid Metabolism: APOH plays a role in the metabolism of lipids, particularly in the transport and clearance of triglyceride-rich lipoproteins such as VLDL and chylomicrons.
  • Coagulation: APOH is involved in the regulation of blood coagulation. It has anticoagulant properties by inhibiting the intrinsic coagulation pathway. It binds to anionic phospholipids on the surfaces of damaged cells and platelets, thereby preventing excessive clot formation.
  • Immune Response: APOH has been implicated in the immune response due to its ability to bind to and neutralize negatively charged phospholipids and lipopolysaccharides. This interaction helps in the clearance of apoptotic cells and bacterial components from the bloodstream.
  • Autoimmune Diseases: APOH is a target antigen in autoimmune conditions such as antiphospholipid syndrome (APS), where autoantibodies against APOH are produced, leading to an increased risk of thrombosis.

Interactions:

  • Lipoprotein Interactions: APOH associates with VLDL and chylomicrons in the blood, aiding in their clearance.
  • Phospholipid Binding: APOH has a strong affinity for anionic phospholipids, which is crucial for its role in coagulation and the immune response.
  • Protein Interactions: It interacts with various plasma proteins, including coagulation factors and complement proteins, modulating their activity.

Clinical Issues:

  • Antiphospholipid Syndrome (APS): APOH is a key autoantigen in APS, a disorder characterized by recurrent thrombosis and pregnancy complications. Autoantibodies against APOH interfere with its anticoagulant function, leading to a hypercoagulable state.
  • Cardiovascular Disease: Due to its role in lipid metabolism and coagulation, altered levels or function of APOH can influence cardiovascular health. Elevated levels of APOH autoantibodies are associated with an increased risk of thrombotic events.
  • Liver Disease: Given that APOH is primarily synthesized in the liver, liver diseases can affect its production and plasma levels, impacting its physiological functions.

Summary:

Apolipoprotein H (APOH) is a multifunctional glycoprotein involved in lipid metabolism, coagulation, and immune response. It circulates in plasma bound to lipoproteins, primarily VLDL and chylomicrons, and plays a critical role in lipid transport and clearance. APOH's ability to bind anionic phospholipids contributes to its anticoagulant and immune regulatory functions. It is a target antigen in antiphospholipid syndrome (APS), where autoantibodies against APOH increase the risk of thrombosis. Understanding APOH's interactions and regulatory mechanisms is vital for developing therapeutic interventions for related disorders, including cardiovascular diseases and autoimmune conditions like APS.
