Description
The urokinase-type plasminogen activator receptor is a key molecule in the regulation of cell-surface plasminogen activation and plays an important role in many normal as well as pathologic processes. The human PLAUR cDNA encodes 335 amino acids including a predicted signal peptide of 22 residues and a hydrophobic C-terminal portion.1 It produces a highly glycosylated protein of about 50 kD in monocytes where it is anchored to the plasma membrane by glycosyl-phosphatidylinositol linkage. PLAUR, also known as UPAR, is directly associated with the carbohydrate-binding domain of SELL in the membrane of neutrophils, an association analogous to that between PLAUR and beta-2 integrins.2 PLAUR-mediated calcium mobilization is SELL dependent. UPAR mRNA levels correlate with the invasive potential of endometrial carcinomas and show a 33-fold increase in UPAR mRNA levels in advanced clinical stage endometrial tumors compared with normal endometrial tissue.3 Furthermore, the increase in UPAR mRNA levels correlated linearly with the progression of disease stage. UPAR protein expressioin correlated positively with rate of recurrence and mortality in patients with endometrial cancer.4 UPAR appears to be a useful prognostic marker for advanced endometrial cancer.

The plasminogen activator, urokinase receptor (PLAUR), also known as urokinase plasminogen activator receptor (uPAR), is a cell surface receptor that plays a pivotal role in modulating extracellular matrix degradation, cell adhesion, and migration. PLAUR is mainly expressed on various cell types, including immune cells (like monocytes and neutrophils), endothelial cells, and cancer cells, and it orchestrates processes crucial for tissue remodeling, inflammation, and metastasis. PLAUR facilitates these processes by binding to urokinase-type plasminogen activator (uPA) and concentrating proteolytic activity on the cell surface, contributing to localized extracellular proteolysis and influencing signal transduction pathways that mediate cell movement and adhesion. This receptor is encoded by the PLAUR gene, located on chromosome 19q13, and belongs to the Ly6/uPAR/α-neurotoxin receptor family, a superfamily of proteins involved in signal transduction.

Urokinase plasminogen activator surface receptor(uPAR, also known as PLAUR or CD87) is a 45-65 kDa, highly glycosylated, GPI-anchored membrane protein. It contains three homologous domains(D1-D3) of which the N-terminal one(D1) represents the uPA-binding domain. After binding to uPAR, urokinase plasminogen activator(uPA) cleaves plasminogen, generating the active protease plasmin which is involved in a wide variety of physiologic and pathologic processes. In addition to regulating proteolysis, uPAR has important function in cell adhesion, migration and proliferation. Studies reveal that uPAR expression is elevated during inflammation and tissue remodelling and in many human cancers, in which it frequently indicates poor prognosis.(PMID 20027185; 12461559)
