UBX domain protein 6 (UBXN6) is a p97/VCP-interacting protein that functions as a key regulator of protein homeostasis by participating in proteasome and autophagy pathways UBXN6 facilitates the extraction and degradation of ubiquitinated misfolded proteins during endoplasmic reticulum-associated degradation (ERAD), playing a vital role in cellular quality control processes It also modulates autophagy by linking p97 to lysosomal pathways for the clearance of protein aggregates Studies show that UBXN6 is involved in cellular stress responses, particularly under proteotoxic conditions where protein turnover mechanisms are critical Dysregulation of UBXN6 leads to protein aggregation and cellular dysfunction, contributing to neurodegenerative diseases such as Parkinson’s and Huntington’s diseases UBXN6 also plays a role in cancer, where its regulation of proteostasis supports tumor cell survival under stress conditions and influences apoptosis Its role in maintaining cellular integrity and proteasome activity highlights its importance in protein quality control and stress adaptation
