UBX domain protein 4 (UBXN4) is a member of the UBX domain-containing protein family that interacts with p97/VCP, a critical ATPase in protein quality control pathways UBXN4 functions as a cofactor that regulates p97 activity during endoplasmic reticulum-associated degradation (ERAD) and the ubiquitin-proteasome system It facilitates the extraction of misfolded proteins from the ER membrane for degradation, maintaining cellular proteostasis under stress conditions UBXN4 is also involved in autophagy and cellular stress responses by coordinating p97-dependent protein turnover Emerging evidence links UBXN4 to DNA damage repair pathways, where it regulates protein stability at replication forks Dysregulation of UBXN4 is associated with neurodegenerative disorders, such as Alzheimer’s disease, where impaired protein clearance results in toxic aggregates Studies also highlight its role in cancer progression, where UBXN4 supports proteasome-mediated protein turnover, contributing to tumor survival and therapy resistance Its ability to regulate proteostasis and stress responses underscores its importance in cellular quality control
