Descripción
TMED3 is a member of the p24 protein family, functioning as a cargo receptor in the early secretory pathway that mediates protein transport between the endoplasmic reticulum (ER), the ER-Golgi intermediate compartment (ERGIC), and the Golgi apparatus. It regulates the selection and packaging of specific cargo proteins into COPII-coated vesicles for export from the ER and coordinates vesicle retrieval via COPI machinery. TMED3 plays an important role in maintaining Golgi structure and the balance of protein flux through the secretory system. Beyond its trafficking role, TMED3 is implicated in Wnt/β-catenin signaling, where it modulates receptor localization and pathway activation critical for cell proliferation, differentiation, and development. Dysregulation of TMED3 contributes to cancers, including colorectal and hepatocellular carcinoma, where altered protein trafficking enhances oncogenic signaling. Knockdown studies reveal impaired vesicular transport, Golgi disorganization, and disrupted Wnt signaling, highlighting its essential role in secretory trafficking, Golgi homeostasis, and signaling regulation.
