SUCNR1, also known as the succinate receptor, is a G protein-coupled receptor that is activated by succinate, an intermediate in the citric acid cycle and a key molecule in cellular metabolism. It primarily couples to Gi proteins, triggering intracellular signaling pathways that regulate vascular tone, inflammation, and metabolic processes. SUCNR1 is expressed in various tissues, including the kidney, vasculature, and immune cells. In the kidney, it plays a role in regulating blood pressure by modulating the renin-angiotensin system. In the vasculature, SUCNR1 is involved in regulating smooth muscle contraction and endothelial cell function, contributing to blood pressure homeostasis. Dysregulation of SUCNR1 has been linked to conditions such as hypertension, ischemia, and inflammation. SUCNR1 is also implicated in cancer progression, particularly in the context of tumor hypoxia. Targeting SUCNR1 with selective agonists or antagonists offers potential therapeutic strategies for managing metabolic diseases, cardiovascular disorders, and inflammation.
