Background

Striatin-3 (STRN3), also known as SG2NA, is a member of the striatin family that functions as a scaffolding protein within the STRIPAK complex. STRN3 coordinates signaling events by interacting with PP2A, kinases, and other regulatory proteins to regulate cell signaling, cytoskeletal dynamics, and vesicular trafficking. It is expressed in various tissues, particularly in the brain, where it modulates calcium-calmodulin signaling pathways essential for neurodevelopment, synaptic plasticity, and neuronal communication. STRN3 plays additional roles in endothelial cell function, angiogenesis, and cell migration through its interactions with signaling proteins. Dysregulation of STRN3 has been implicated in cancers and neurological disorders, where altered PP2A activity and disrupted signaling pathways contribute to disease progression. Functional studies reveal that loss of STRN3 impairs cellular signaling, cytoskeletal reorganization, and vesicle transport, emphasizing its role in cellular communication, cytoskeletal integrity, and signaling homeostasis.
