Descripción

S1PR4 is a G protein-coupled receptor for sphingosine-1-phosphate (S1P), primarily expressed in hematopoietic cells, including lymphocytes, dendritic cells, and natural killer (NK) cells. S1PR4 couples to Gi and G12/13 proteins, regulating pathways like PI3K/Akt and Rho/ROCK, which control cell migration, immune responses, and cytoskeletal reorganization. S1PR4 plays a critical role in immune cell trafficking, influencing lymphocyte egress from lymphoid tissues and modulating inflammatory responses. It is involved in regulating dendritic cell maturation and T-cell function, positioning it as a key player in adaptive immunity. Dysregulation of S1PR4 is associated with autoimmune diseases, inflammatory disorders, and hematological malignancies. In cancer, S1PR4 has been linked to tumor progression and immune evasion, particularly in leukemia and lymphoma. S1PR4 antagonists are being explored for therapeutic use in autoimmune diseases and cancers by modulating immune cell function and migration, highlighting its importance in immune regulation.
