Descripción

LRRC8E is a subunit of the volume-regulated anion channel (VRAC), which forms heteromeric channels in combination with LRRC8A and other LRRC8 paralogs to regulate chloride and osmolyte efflux under osmotic stress. LRRC8E contributes to the fine-tuning of VRAC conductance, ion selectivity, and channel kinetics, ensuring cellular adaptation to hypotonic conditions and mechanical stress. It is expressed in various tissues, including immune cells, muscle, and neuronal tissues, where it regulates cell volume, apoptosis, and ionic homeostasis. LRRC8E plays a role in immune cell function, proliferation, and cellular responses to swelling-induced stress, participating in processes like immune signaling and apoptotic volume decrease. Dysregulation of LRRC8E disrupts VRAC activity, contributing to pathological conditions such as immune dysfunction, neurodegenerative diseases, and cancer, where impaired ion homeostasis affects cell survival and growth. Knockout studies reveal diminished swelling-induced chloride currents, reduced apoptotic responses, and impaired cell volume regulation, underscoring its essential role in osmoregulation and adaptive responses to environmental stress.
