Descripción

TMED7, a key p24 family protein, regulates the trafficking of proteins and receptors through the early secretory pathway, connecting the ER, ER-Golgi intermediate compartment (ERGIC), and Golgi apparatus. It functions as a cargo receptor, enabling the export of correctly folded proteins in COPII-coated vesicles and mediating their retrieval via COPI. TMED7 also influences receptor signaling and immune responses, particularly through its involvement in toll-like receptor (TLR) pathways, where it modulates TLR4 trafficking and downstream activation of pro-inflammatory signaling cascades. TMED7 is widely expressed across immune cells and secretory tissues, where it supports vesicular transport efficiency, receptor localization, and cytokine production. Dysregulation of TMED7 is implicated in immune disorders, neurodegeneration, and cancers, where disrupted protein trafficking and receptor signaling contribute to disease pathogenesis. Knockdown studies reveal reduced TLR signaling, impaired vesicle formation, and increased ER stress, underscoring its role in vesicle trafficking, immune signaling, and cellular homeostasis.
