Acetyl Coenzyme A Acyltransferase 1A (ACAA1A) is a peroxisomal enzyme that plays a critical role in the beta-oxidation pathway, facilitating the breakdown of long-chain and branched-chain fatty acids into acetyl-CoA and shorter acyl-CoA molecules. ACAA1A functions as a thiolase, catalyzing the final step in fatty acid degradation, which is essential for lipid homeostasis and energy production, particularly during fasting or other conditions where fatty acids serve as the primary energy source. ACAA1A is highly expressed in tissues with active lipid metabolism, including the liver, kidneys, and adipose tissue. Mutations or deficiencies in ACAA1A activity can lead to peroxisomal disorders, such as Zellweger syndrome or adrenoleukodystrophy, characterized by the accumulation of very-long-chain fatty acids and associated metabolic dysfunctions. ACAA1A is also of interest in studies on metabolic reprogramming in cancer, as its role in fatty acid oxidation may influence tumor growth and progression.
