Ragulator complex protein LAMTOR1 is a core component of the Ragulator complex, which anchors Rag GTPases to the lysosomal membrane, functioning as a key regulator of mTORC1 signaling LAMTOR1 is essential for nutrient sensing and the activation of mTORC1, a central signaling hub that regulates cellular metabolism, growth, and autophagy in response to amino acid availability It interacts with other components of the Ragulator complex to facilitate lysosome-localized mTORC1 activation, ensuring proper coordination of anabolic and catabolic processes Dysregulation of LAMTOR1 impairs mTORC1 signaling, contributing to metabolic disorders, cancer progression, and neurodegenerative diseases where nutrient sensing and autophagic balance are disrupted LAMTOR1 is also involved in lysosomal positioning and function, highlighting its role in cellular homeostasis and stress responses Recent studies have demonstrated its involvement in immune cell activation and pathogen response, further emphasizing its critical function as a central regulator of lysosome-mTORC1 signaling and cellular metabolism
