BRICK1 subunit of SCAR/WAVE actin nucleating complex (BRK1) is a protein that is a part of the SCAR/WAVE complex, which regulates actin polymerization by activating the Arp2/3 complex The SCAR/WAVE complex, including BRICK1, is involved in initiating the formation of branched actin filaments, a process essential for cell movement, membrane ruffling, and cellular shape changes BRICK1 plays a role in promoting actin dynamics, particularly in immune cells where it influences processes such as T-cell activation, cell migration, and phagocytosis In addition, BRICK1 is important for neuronal development, where it helps regulate synaptic plasticity and dendritic spine morphology Disruption of BRICK1 or the SCAR/WAVE complex can lead to defects in cell motility, immune function, and nervous system development Abnormal regulation of actin dynamics due to BRICK1 dysfunction is implicated in various diseases, including autoimmune disorders, cancer metastasis, and neurodevelopmental disorders where impaired cellular movement and actin remodeling contribute to disease progression
