Descripción

ALOX15 catalyzes the oxygenation of arachidonic acid and linoleic acid to generate 15(S)-hydroperoxyeicosatetraenoic acid [15(S)-HPETE] and 13(S)-hydroxyoctadecadienoic acid (13(S)-HODE), respectively. These lipid intermediates are precursors for resolvins, protectins, and other specialized pro-resolving mediators (SPMs) involved in the resolution of inflammation. ALOX15 is highly expressed in reticulocytes, macrophages, and epithelial cells, where it plays roles in redox regulation, membrane remodeling, and anti-inflammatory responses. In macrophages, ALOX15 promotes the clearance of apoptotic cells (efferocytosis) and contributes to tissue homeostasis during inflammation resolution. Dysregulation of ALOX15 is associated with various pathological conditions, including asthma, atherosclerosis, diabetes, and neurodegenerative diseases. In cancer biology, ALOX15 has dual roles, acting as both a tumor suppressor by inhibiting proliferation and a pro-tumorigenic factor in specific contexts. Its ability to generate lipid mediators essential for resolving inflammation highlights its therapeutic potential in chronic inflammatory and metabolic disorders.
