Description

This gene encodes the B-cell lineage specific activator protein that is expressed at early, but not late stages of B-cell differentiation. Its expression has also been detected in developing CNS and testis and so the encoded protein may also play a role in neural development and spermatogenesis. This gene is located at 9p13, which is involved in t (9;14) (p13;q32) translocations recurring in small lymphocytic lymphomas of the plasmacytoid subtype, and in derived large-cell lymphomas. This translocation brings the potent E-mu enhancer of the IgH gene into close proximity of the PAX5 promoter, suggesting that the deregulation of transcription of this gene contributes to the pathogenesis of these lymphomas. Alternatively spliced transcript variants encoding different isoforms have been described but their biological validity has not been determined. Descripción

PAX5 is a critical transcription factor that governs B-cell lineage commitment, differentiation, and survival during hematopoiesis. PAX5 activates B-cell-specific gene expression while repressing alternative lineage programs to ensure B-cell identity. It is expressed in pro-B cells and remains active throughout B-cell maturation, where it regulates immunoglobulin gene rearrangement, signaling, and antigen response. Dysregulation or mutations in PAX5 are commonly observed in B-cell acute lymphoblastic leukemia (B-ALL), where its loss contributes to impaired B-cell differentiation and leukemogenesis. PAX5 also participates in neural and midbrain development during embryogenesis. Knockout models show arrested B-cell development at the pro-B stage and defects in midbrain formation, highlighting its role in both hematopoiesis and neurodevelopment. In cancer, altered PAX5 expression disrupts immune regulation, contributes to lymphomagenesis, and serves as a biomarker for diagnosing B-cell malignancies.
