Nuclear receptor subfamily 4 group A member 3 (NR4A3), also known as NOR1, is a nuclear receptor that regulates gene expression in response to various extracellular signals, including growth factors, stress hormones, and cytokines NR4A3 is involved in controlling cell proliferation, apoptosis, and differentiation, particularly in immune cells and tissues related to metabolism It plays a role in regulating inflammation and immune responses by modulating the expression of pro-inflammatory genes, thus influencing conditions such as atherosclerosis, diabetes, and autoimmune diseases NR4A3 also has functions in metabolic processes, including the regulation of lipid metabolism and insulin sensitivity, and is involved in the response to oxidative stress and cellular damage Dysregulation of NR4A3 has been linked to various pathological conditions, including cardiovascular diseases, cancer, and chronic inflammatory diseases It also plays a role in regulating the circadian rhythm and metabolic health, highlighting its potential as a target for therapeutic strategies aimed at treating metabolic and immune-related disorders

This gene encodes a member of the steroid-thyroid hormone-retinoid receptor superfamily. The encoded protein may act as a transcriptional activator. The protein can efficiently bind the NGFI-B Response Element (NBRE). Three different versions of extraskeletal myxoid chondrosarcomas (EMCs) are the result of reciprocal translocations between this gene and other genes. The translocation breakpoints are associated with Nuclear Receptor Subfamily 4, Group A, Member 3 (on chromosome 9) and either Ewing Sarcome Breakpoint Region 1 (on chromosome 22), RNA Polymerase II, TATA Box-Binding Protein-Associated Factor, 68-KD (on chromosome 17), or Transcription factor 12 (on chromosome 15). Multiple transcript variants encoding different isoforms have been found for this gene.
