Neurogenic locus notch homolog protein 4 (NOTCH4) is a transmembrane receptor in the Notch family, crucial for cell-cell signaling in endothelial cells, where it regulates angiogenesis, vascular remodeling, and homeostasis It encodes a single-pass transmembrane protein with extracellular EGF-like repeats for ligand binding and an intracellular domain (NICD) that translocates to the nucleus upon activation Binding to ligands like Delta-like (DLL) or Jagged triggers proteolytic cleavages by ADAM proteases and gamma-secretase, releasing NICD to activate target genes such as HES and HEY, which control cell proliferation, differentiation, and apoptosis Dysregulation of NOTCH4 signaling is linked to cancers, particularly breast cancer and gliomas, due to its role in angiogenesis and metastasis It also modulates endothelial inflammation and immune responses Mutations or altered expression of NOTCH4 contribute to vascular abnormalities like arteriovenous malformations and pulmonary hypertension Genetic variations in NOTCH4 are associated with schizophrenia and autoimmune diseases like systemic lupus erythematosus, highlighting its importance in development and pathology
