NCK associated protein 1 (NCKAP1) is an actin-regulatory protein that interacts with NCK adapter proteins and plays a crucial role in regulating cytoskeletal dynamics in various cell types NCKAP1 activates the Arp2/3 complex to promote the formation of branched actin filaments, facilitating the movement of cells and the establishment of cellular polarity This function is vital for processes like cell migration, adhesion, and tissue development, particularly in immune cells such as T cells and macrophages, where it regulates key immune responses such as synapse formation and phagocytosis NCKAP1 also contributes to the remodeling of the cytoskeleton in neurons, where it affects synaptic function and plasticity, which are critical for learning and memory However, alterations in NCKAP1 expression or function have been linked to a range of diseases, including immune disorders, cancer metastasis, and neurodevelopmental disorders Abnormal actin dynamics due to NCKAP1 dysregulation can impair immune cell function and facilitate the invasive capabilities of cancer cells, making it an important target for therapeutic strategies aimed at treating inflammatory diseases and controlling cancer metastasis
