Zinc Finger MIZ-Type Containing 2 (ZMIZ2) is a transcriptional co-activator of the PIAS-like family, containing a MIZ domain that mediates interactions with transcription factors and chromatin regulators It enhances the transcriptional activity of androgen receptors (AR) and interacts with pathways such as p53, SMAD, and Wnt signaling ZMIZ2 contributes to essential cellular processes including proliferation, differentiation, DNA repair, and hematopoietic stem cell regulation Its role in immune cell function and T-cell activation highlights its importance in immune homeostasis Dysregulation of ZMIZ2 expression or mutations has been linked to cancers including prostate, breast, and colorectal cancers, where it can promote oncogenic transcriptional activity and enhance tumor progression ZMIZ2 has also been implicated in autoimmune diseases such as lupus and multiple sclerosis due to its involvement in immune tolerance and inflammatory responses Emerging studies suggest its function in modulating chromatin accessibility and epigenetic regulation, making it a key player in transcriptional regulation and cellular homeostasis
