Descripción
DENN domain containing 6A (DENND6A) is a member of the DENN domain family, acting as a guanine nucleotide exchange factor (GEF) for Rab GTPases, specifically Rab14. DENND6A regulates intracellular trafficking, including vesicle transport and endosomal recycling, by promoting the GDP-to-GTP exchange on Rab14. This activity is essential for processes like endocytic recycling of plasma membrane proteins, intracellular signaling, and maintenance of epithelial cell polarity. DENND6A also supports trafficking of cargo between endosomes and the Golgi apparatus, which is critical for protein sorting and secretion. Its expression is ubiquitous, with heightened activity in polarized cells such as neurons and epithelial tissues. Dysregulation of DENND6A has been linked to defects in intracellular transport, contributing to diseases such as cancer and neurological disorders, where disruptions in endosomal sorting and recycling lead to altered cellular signaling and polarity. DENND6A’s role in vesicle dynamics highlights its importance in maintaining cellular organization and adaptive responses.
