Descripción

PLEKHA2 is a lipid-binding protein that contains a pleckstrin homology (PH) domain with high specificity for phosphoinositides such as PI(3,4)P2, enabling it to localize to plasma membranes and mediate signaling events. It plays a key role in actin cytoskeletal dynamics, cellular migration, and immune cell signaling by recruiting cytoskeletal regulators and signaling molecules to membrane sites. PLEKHA2 is expressed in immune and endothelial cells, where it regulates processes such as phagocytosis, chemotaxis, and membrane ruffling during immune responses. It also participates in signaling pathways like PI3K/Akt, contributing to cell survival and proliferation. Dysregulation of PLEKHA2 has been associated with inflammatory diseases and cancer metastasis due to altered actin remodeling and immune cell dysfunction. Knockout studies demonstrate impaired actin polymerization, reduced immune cell migration, and defects in membrane organization, highlighting its role as a mediator of phosphoinositide-driven cellular responses.
