Descripción
GPR18 is an orphan G protein-coupled receptor implicated in immune regulation, lipid signaling, and cannabinoid-like responses. It is widely expressed in immune cells, the central nervous system, and peripheral tissues. GPR18 is activated by endogenous ligands, such as N-arachidonoylglycine (NAGly), and interacts with cannabinoid-like signaling pathways. GPR18 regulates immune cell migration, particularly in macrophages, microglia, and T cells, where it modulates inflammation and resolution of tissue injury. It has been linked to ocular pressure regulation in the eye, making it a potential target for treating glaucoma. In cancer, GPR18 expression promotes tumor progression, survival, and metastasis, particularly in melanoma and breast cancer, where it enhances cell migration and immune evasion. Dysregulation of GPR18 signaling has also been associated with inflammatory and neurodegenerative diseases. Its involvement in immune homeostasis and cannabinoid-like signaling makes GPR18 a promising target for therapeutic development in inflammation, cancer, and neuroimmune disorders.
