Descripción
LRRC8A is a core component of the volume-regulated anion channel (VRAC), a heteromeric channel responsible for regulating cell volume through the efflux of chloride ions and osmolytes under hypotonic stress. It contains leucine-rich repeats (LRRs) and transmembrane domains, which enable it to form complexes with other LRRC8 family members, such as LRRC8B-E, to assemble functional VRAC channels. LRRC8A serves as an essential subunit for channel formation, as it interacts with partner subunits to determine VRAC conductance, ion selectivity, and tissue-specific function. It plays critical roles in cell volume regulation, apoptosis, and osmotic homeostasis, particularly in response to swelling-induced mechanical stress. LRRC8A is widely expressed across tissues, including epithelial cells, immune cells, and neurons. Dysregulation of LRRC8A impairs volume regulation, contributing to pathological conditions such as neurodegenerative diseases, ischemia, and cancer progression due to defective apoptotic signaling and cellular homeostasis. Knockout studies reveal impaired VRAC activity, compromised cell survival under osmotic stress, and defects in apoptosis, highlighting its importance in maintaining cellular ion balance and volume regulation.
