DENN domain containing 1B (DENND1B) is a member of the DENN domain protein family, serving as a guanine nucleotide exchange factor (GEF) for Rab GTPases, specifically activating Rab35. Through its conserved DENN domain, DENND1B regulates endosomal trafficking and recycling processes, ensuring the sorting, transport, and fusion of vesicles within the endosomal system. It plays a critical role in cytokinesis by facilitating plasma membrane recycling and the proper separation of daughter cells during cell division. DENND1B is also implicated in immune cell signaling, where it contributes to cytokine secretion and receptor recycling in inflammatory responses. Dysregulation of DENND1B has been linked to increased susceptibility to inflammatory diseases, including asthma, due to its role in controlling immune signaling pathways. Its expression in immune-related tissues underscores its importance in coordinating immune cell responses and maintaining intracellular transport mechanisms. DENND1B also interacts with other Rab GTPases, further emphasizing its versatility in regulating vesicular dynamics and membrane remodeling.
