KCTD16 is another member of the KCTD family that functions as an auxiliary subunit of GABA-B receptors, modulating their pharmacological and signaling properties. KCTD16 specifically regulates the kinetics of GABA-B receptor activation and desensitization, influencing inhibitory neurotransmission in the central nervous system. It is widely expressed in brain regions associated with cognition, motor activity, and emotional regulation, playing a role in synaptic plasticity and neuronal excitability. KCTD16 interacts with GABA-B receptor subunits to enhance receptor sensitivity and prolong inhibitory signaling, which is critical for maintaining neural network stability. Dysregulation of KCTD16 has been linked to psychiatric disorders, including schizophrenia, anxiety, and depression, where impaired GABA-B receptor function contributes to disrupted inhibitory signaling. Beyond the nervous system, KCTD16 may participate in protein degradation pathways through its BTB/POZ domain, influencing cellular homeostasis. Its ability to fine-tune GABA-B receptor signaling positions KCTD16 as a key target for therapeutic interventions aimed at restoring inhibitory balance in neurological and psychiatric disorders.
