Descripción
KCTD12 is a member of the potassium channel tetramerization domain (KCTD) family and functions as an auxiliary subunit of GABA-B receptors. KCTD12 modulates GABA-B receptor signaling by accelerating activation and desensitization kinetics, fine-tuning inhibitory neurotransmission in the central nervous system. It is predominantly expressed in brain regions involved in cognition, sensory processing, and motor control, where it regulates synaptic inhibition and neuronal excitability. KCTD12 also interacts with various proteins through its BTB/POZ domain, contributing to cellular processes like ubiquitination and transcriptional regulation. Beyond its role in neurotransmission, KCTD12 has been implicated in cancer, particularly gastrointestinal stromal tumors (GISTs), where its expression influences tumor cell proliferation and survival. In psychiatric disorders, such as anxiety and depression, altered KCTD12 expression disrupts GABA-B receptor-mediated signaling, impairing inhibitory balance in neural circuits. Its role as a GABA-B receptor modulator and involvement in protein-protein interactions highlights KCTD12 as a key regulator of neuronal function and a potential therapeutic target for neurological disorders and cancer.
