ARFGEF family member 3 (ARFGEF3) is another guanine nucleotide exchange factor that activates ARF proteins, playing a crucial role in regulating intracellular trafficking and actin cytoskeleton organization ARFGEF3 works by catalyzing the exchange of GDP for GTP on ARF proteins, enabling ARF-mediated signaling and regulating processes like vesicular trafficking, membrane dynamics, and cell signaling It is involved in cellular events such as endocytosis, exocytosis, and the formation of membrane-bound organelles ARFGEF3 also influences actin dynamics, which are essential for cell movement, adhesion, and shape maintenance Additionally, ARFGEF3 has been linked to processes in the immune system, particularly in the regulation of immune cell activation and trafficking Disruption in ARFGEF3 activity has been associated with various diseases, including cancer, where altered cell trafficking and actin dynamics contribute to metastasis and tumor progression It also plays a role in other conditions such as neurodegenerative diseases, where disruptions in cellular trafficking and signaling may contribute to cellular dysfunction
