ABCB9, also known as TAPL (Transporter Associated with Antigen Processing-Like), is an ATP-binding cassette transporter localized to lysosomes and late endosomes. It plays a role in the transport of peptides into lysosomal compartments, contributing to processes such as antigen presentation and lysosomal proteostasis. ABCB9 contains two nucleotide-binding domains and transmembrane regions that mediate ATP-dependent peptide transport. It is expressed in various tissues, with higher levels in immune-related cells, where it supports lysosomal function and immune responses. Although its precise physiological substrates and roles remain under investigation, ABCB9’s involvement in lysosomal function underscores its importance in cellular homeostasis, immune regulation, and protein degradation pathways.

This protein is a member of the MDR/TAP subfamily. Members of the MDR/TAP subfamily are involved in multidrug resistance as well as antigen presentation. This family member functions in the translocation of peptides from the cytosol into the lysosomal lumen. Alternative splicing of this gene results in distinct isoforms which are likely to have different substrate specificities.
