ALKBH8 is a unique member of the ALKB family that functions as a tRNA methyltransferase and oxidative repair enzyme. It catalyzes the methylation of specific nucleotides in tRNA, particularly wobble uridine residues, ensuring proper tRNA stability, codon recognition, and accurate translation. ALKBH8 also possesses RNA repair activity, where it oxidatively repairs methylated lesions in tRNA, ensuring the maintenance of functional tRNA pools. This dual function makes ALKBH8 critical for maintaining translational fidelity and cellular protein synthesis. ALKBH8 is particularly important under oxidative stress, where it repairs alkylation-induced RNA damage and prevents translational errors. Dysregulation of ALKBH8 has been linked to cancer progression, as defective tRNA modification can disrupt protein synthesis and cellular homeostasis. Its role in enhancing translational accuracy and stress adaptation highlights its importance in tumor suppression, metabolic regulation, and cellular defense mechanisms.
