Descripción

ACER2 is a membrane-bound ceramidase that hydrolyzes a variety of ceramide species into sphingosine and fatty acids under alkaline conditions, regulating sphingolipid metabolism across diverse cellular contexts. ACER2 is expressed in numerous tissues, including the liver, brain, immune cells, and lungs, reflecting its broad functional roles. It is critical for generating sphingosine, which is phosphorylated by sphingosine kinases to produce sphingosine-1-phosphate (S1P), a bioactive lipid that mediates cell survival, migration, inflammation, and angiogenesis. ACER2 also contributes to immune system regulation, particularly in lymphocyte trafficking and maintaining vascular integrity. Dysregulation of ACER2 activity has been linked to diseases such as neurodegeneration, metabolic disorders, and cancer progression, where altered ceramide and S1P levels disrupt cell signaling. Emerging evidence highlights its role in apoptotic pathways and chemotherapy resistance in cancer cells, further emphasizing its therapeutic potential as a target for modulating sphingolipid metabolism and associated signaling pathways.
