Background

ACER2 is a membrane-bound ceramidase that hydrolyzes a variety of ceramide species into sphingosine and fatty acids under alkaline conditions, regulating sphingolipid metabolism across diverse cellular contexts. ACER2 is expressed in numerous tissues, including the liver, brain, immune cells, and lungs, reflecting its broad functional roles. It is critical for generating sphingosine, which is phosphorylated by sphingosine kinases to produce sphingosine-1-phosphate (S1P), a bioactive lipid that mediates cell survival, migration, inflammation, and angiogenesis.
