LPAR6, also known as P2Y5, is a G protein-coupled receptor activated by lysophosphatidic acid (LPA). It primarily couples to G12/13 proteins and plays a key role in cellular signaling related to cytoskeletal reorganization, cell migration, and proliferation. LPAR6 is predominantly expressed in hair follicles, skin, and epithelial tissues, where it regulates hair growth and skin homeostasis. Mutations in LPAR6 are associated with inherited hair disorders such as hypotrichosis and woolly hair syndrome, where impaired LPA signaling disrupts normal hair follicle function. Beyond its role in hair biology, LPAR6 contributes to epithelial cell survival and wound healing processes. LPAR6 has also been linked to cancer progression, particularly in promoting cell migration, metastasis, and resistance to apoptosis in epithelial cancers. Dysregulation of LPAR6 signaling has implications in skin disorders, inflammatory conditions, and tumor progression. LPAR6 represents a therapeutic target for managing hair growth disorders, epithelial repair, and cancer metastasis.
