Descripción
LRRC8C is a subunit of the volume-regulated anion channel (VRAC), where it assembles with LRRC8A and other LRRC8 paralogs to form functional heteromeric channels involved in chloride and osmolyte efflux under osmotic stress. LRRC8C modulates the biophysical properties of VRAC, such as ion selectivity and conductance, determining the channel's responsiveness to hypotonic swelling and cellular needs. It is expressed across multiple tissues, including epithelial cells, muscle, and immune cells, where it regulates cell volume, proliferation, and apoptosis. LRRC8C is critical for osmoregulation, immune responses, and apoptotic pathways, particularly during cellular swelling and mechanical stress. Dysregulation of LRRC8C disrupts ion homeostasis, leading to pathological conditions such as immune dysregulation, neurodegeneration, and tumor progression due to defective apoptosis and cell volume control. Knockout models exhibit impaired VRAC activity, altered chloride efflux, and reduced apoptosis under osmotic stress, underscoring its role in maintaining ionic balance, cell survival, and adaptive cellular responses.
