Descripción

ADGRL3, also referred to as Latrophilin-3, is primarily expressed in the central nervous system and plays a pivotal role in neural circuit formation, synaptic development, and plasticity. Its interaction with neuronal ligands facilitates trans-synaptic adhesion, critical for the regulation of synaptic strength and maintenance of neural connectivity. ADGRL3 is implicated in neuropsychiatric and developmental disorders, with genetic mutations linked to conditions such as attention-deficit/hyperactivity disorder (ADHD) and autism spectrum disorders. It regulates intracellular signaling pathways involved in neuronal differentiation and synaptic maturation, underscoring its role in both developmental and functional aspects of the nervous system. Dysregulation of ADGRL3 can lead to altered neuronal communication, impacting behavior, cognition, and overall neurological health.

This gene encodes a member of the latrophilin subfamily of G-protein coupled receptors (GPCR). Latrophilins may function in both cell adhesion and signal transduction. In experiments with non-human species, endogenous proteolytic cleavage within a cysteine-rich GPS (G-protein-coupled-receptor proteolysis site) domain resulted in two subunits (a large extracellular N-terminal cell adhesion subunit and a subunit with substantial similarity to the secretin/calcitonin family of GPCRs) being non-covalently bound at the cell membrane.
