IL10RA is a critical component of the interleukin-10 (IL10) receptor complex, primarily expressed on immune cells like monocytes, macrophages, dendritic cells, and T cells This receptor binds IL10, a potent anti-inflammatory cytokine, and works in conjunction with IL10 receptor beta (IL10RB) to mediate downstream signaling through the JAK1/STAT3 pathway IL10RA contains an extracellular domain that facilitates IL10 binding, a transmembrane region, and an intracellular segment that interacts with JAK1 kinase Activation of IL10RA suppresses pro-inflammatory cytokines, including TNF-α, IL6, and IL12, while enhancing immune tolerance and promoting tissue homeostasis Loss-of-function mutations in IL10RA disrupt IL10 signaling, leading to early-onset inflammatory bowel disease (IBD) and other severe inflammatory conditions, often in childhood IL10RA dysfunction results in an inability to regulate immune responses effectively, leading to hyperactivation of inflammatory pathways Therapies targeting IL10RA, such as recombinant IL10 or receptor-modulating agents, are being investigated for autoimmune diseases and inflammatory disorders due to their ability to restore IL10-mediated anti-inflammatory signaling IL10RA also plays a role in tumor progression by suppressing immune activation in the tumor microenvironment, making it a potential immunotherapy target

It has been shown to mediate the immunosuppressive signal of interleukin 10, and thus inhibits the synthesis of proinflammatory cytokines.
