Background

YWHAB, part of the 14-3-3 protein family, functions as a scaffold protein that binds to phosphorylated serine/threonine-containing motifs, regulating protein localization, stability, and signaling pathways. It is involved in processes such as cell cycle control, DNA damage response, apoptosis, and stress adaptation by interacting with key signaling proteins, including Raf kinases, BAD, and CDC25. YWHAB regulates pathways like MAPK and PI3K/Akt, influencing cellular growth, survival, and metabolism. It is ubiquitously expressed, with high levels in the brain, where it contributes to synaptic plasticity, neuroprotection, and neuronal development. Dysregulation of YWHAB has been associated with neurodegenerative diseases, including Alzheimer’s and Parkinson’s, as well as cancer, where altered protein interactions promote tumor survival and metastasis.
