Descripción
SH2D4B is an SH2 domain-containing adaptor protein that functions as a modulator of cytokine and receptor tyrosine kinase signaling pathways. It negatively regulates signaling cascades such as JAK/STAT and PI3K/Akt by binding to phosphotyrosine residues on activated receptors or signaling intermediates, attenuating downstream signaling. SH2D4B is expressed in various tissues, including hematopoietic and epithelial cells, where it controls cellular proliferation, differentiation, and immune responses. It acts as a tumor suppressor by inhibiting excessive signaling that promotes cell survival and uncontrolled proliferation. Dysregulation or loss of SH2D4B expression is associated with cancer progression and immune dysfunction due to hyperactive cytokine signaling and impaired negative feedback regulation. Knockdown studies reveal increased cell proliferation, altered cytokine responses, and enhanced tumor growth, highlighting its role as a critical negative regulator of signal transduction and cellular homeostasis.
