SAM and SH3 domain containing 3 (SASH3), also known as the signaling lymphocyte activation molecule (SLAM)-associated protein-like molecule, is an adaptor protein predominantly expressed in immune cells, including T cells and B cells SASH3 plays a critical role in T-cell receptor (TCR) and B-cell receptor (BCR) signaling, where it facilitates downstream pathways essential for lymphocyte activation, differentiation, and survival Through its SH3 and SAM domains, SASH3 interacts with other signaling proteins and regulates immune responses, including cytokine production and antigen recognition Dysregulation or mutations in SASH3 are linked to immune deficiencies and autoimmune diseases, as impaired signaling affects lymphocyte function and immune tolerance SASH3 is also implicated in hematopoietic malignancies, where its altered expression contributes to abnormal immune cell proliferation and survival Recent studies indicate its role in regulating inflammation and adaptive immunity, highlighting its importance in maintaining immune system balance Its involvement in critical signaling pathways positions SASH3 as a key regulator of immune function and a potential target for therapeutic intervention in immunological disorders and cancers
