Descripción
PLEKHA7 is a pleckstrin homology (PH) domain-containing protein that localizes at adherens junctions in epithelial cells, where it plays a critical role in maintaining cell-cell adhesion and epithelial polarity. It interacts with junctional proteins like E-cadherin and components of the cytoskeleton to stabilize cell junctions, supporting tissue organization and barrier integrity. PLEKHA7 is involved in the regulation of Rho GTPase activity, influencing actin cytoskeleton dynamics and cell signaling pathways that control adhesion and migration. It is widely expressed in epithelial tissues, particularly in organs such as the intestine and kidney, where it contributes to tissue homeostasis and repair. Dysregulation or loss of PLEKHA7 has been associated with cancer progression, where disrupted cell junctions promote epithelial-mesenchymal transition (EMT), tumor invasion, and metastasis. Knockdown studies reveal impaired adherens junctions, reduced epithelial polarity, and increased cell motility, highlighting its essential role in maintaining epithelial integrity and suppressing tumorigenesis.
