Background

P2RY8 is an orphan G protein-coupled receptor within the P2Y receptor family, presumed to respond to extracellular nucleotides like ATP and UDP, although its endogenous ligand remains unclear. P2RY8 is expressed predominantly in immune cells, particularly in lymphocytes and germinal centers of lymphoid tissues, where it modulates immune signaling, cell migration, and lymphocyte development. Its role in immune function positions P2RY8 as a critical regulator of adaptive immunity and germinal center reactions. Dysregulation of P2RY8 signaling has been implicated in hematological malignancies, including B-cell lymphomas and acute lymphoblastic leukemia (ALL), where P2RY8 rearrangements are associated with impaired lymphocyte differentiation and tumor progression. The exact signaling mechanisms and downstream pathways of P2RY8 remain under investigation, but it is emerging as a potential biomarker and therapeutic target in immune-related cancers.
