Descripción
NPBWR2, also known as GPR8, is a G protein-coupled receptor with high affinity for neuropeptide B (NPB) and neuropeptide W (NPW). Unlike NPBWR1, NPBWR2 expression is more limited to specific regions of the brain and peripheral tissues, particularly in the hypothalamus, thalamus, and spinal cord. NPBWR2 couples to Gi/o proteins, inhibiting adenylate cyclase, reducing cAMP levels, and modulating cellular excitability and neurotransmitter release. NPBWR2 is involved in the regulation of feeding behavior, stress responses, and energy metabolism, similar to NPBWR1. It plays a role in modulating nociception and pain perception, particularly in the spinal cord, where it regulates pain transmission. Dysregulation of NPBWR2 has been associated with metabolic disorders, stress-related conditions, and chronic pain syndromes. NPBWR2 is being explored as a therapeutic target for obesity, mood disorders, and pain management, as it influences energy balance, emotional regulation, and nociceptive pathways. Its selective expression and functional roles make it a promising candidate for targeted therapies in CNS disorders.
