Background

Lysosome-associated membrane glycoprotein 5 (LAMP5) is a member of the LAMP family of lysosomal membrane proteins, primarily expressed in neuronal tissues and specific cell types such as inhibitory interneurons LAMP5 plays a key role in maintaining lysosomal function, particularly in neurons, where it regulates endosomal-lysosomal trafficking and protein degradation pathways It is involved in synaptic transmission and neuronal homeostasis, contributing to proper neuronal development and function Dysregulation of LAMP5 expression has been associated with neurodevelopmental disorders and neurodegenerative diseases such as Alzheimer’s disease and epilepsy, where impaired lysosomal activity leads to neuronal dysfunction LAMP5 also plays a role in autophagy and cellular stress responses, where it regulates lysosomal biogenesis and fusion processes Recent studies have highlighted its potential involvement in tumorigenesis, particularly in brain cancers, where it can influence lysosome-mediated processes critical for tumor progression and survival
