Descripción

LRRC8B is a subunit of the volume-regulated anion channel (VRAC) that partners with LRRC8A to form heteromeric channels regulating the efflux of chloride ions and osmolytes during cell swelling and osmotic stress. It contains leucine-rich repeats (LRRs) that facilitate protein-protein interactions, modulating VRAC properties such as ion permeability and channel kinetics. LRRC8B is expressed in a variety of tissues, including the brain, kidney, and immune cells, where it contributes to cellular osmoregulation, apoptosis, and volume homeostasis. It plays a role in the fine-tuning of VRAC activity, influencing chloride conductance and transport specificity depending on cellular and environmental conditions. Dysregulation of LRRC8B has been associated with impaired cell volume control, altered apoptotic responses, and cellular dysfunction under osmotic stress, contributing to diseases like inflammation, neurodegeneration, and cancer. Knockout studies reveal diminished VRAC activity, reduced swelling-induced ion efflux, and impaired cellular adaptation to hypotonic stress, highlighting its role in maintaining cellular volume and ionic balance.
