IL10RB is a shared receptor subunit within the IL10 receptor complex that interacts with IL10RA to propagate IL10 signaling It is widely expressed across immune and non-immune cell types, highlighting its role in diverse biological processes beyond IL10 signaling IL10RB forms a functional receptor complex with IL10RA and is also involved in the signaling pathways of cytokines such as IL22, IL26, and type III interferons (IFN-λ) Upon binding IL10, IL10RB recruits TYK2 kinase, facilitating activation of the JAK1/STAT3 signaling cascade to suppress pro-inflammatory cytokines and regulate immune tolerance Mutations in IL10RB lead to disrupted IL10 signaling and are strongly linked to severe early-onset IBD, systemic inflammation, and immune dysregulation IL10RB’s shared use in other cytokine pathways highlights its broad immunoregulatory function, making it crucial in the resolution of inflammation and antiviral immune responses Dysregulation of IL10RB has been associated with chronic inflammatory disorders, autoimmunity, and heightened susceptibility to viral infections Therapeutic approaches aimed at modulating IL10RB activity are being explored for managing inflammatory and infectious diseases, as enhancing IL10RB signaling can reestablish immune balance and prevent tissue damage caused by excessive inflammation
